1 Scope This part of GB/T 18204 specifies the on-site sampling and laboratory cultivation methods of total bacterial count, total fungi count, β-hemolytic streptococcus and legionella pneumophila in air of public places. This part is applicable to the determination of total bacterial count, total fungi count, β-hemolytic streptococcus and legionella pneumophila in air of public places; the determination of these in other places is implemented by reference to this part. Note: if two or more examination methods are available for the same index in this part, specific method may be selected according to technical conditions. 2 Terms and Definitions For the purposes of this document, the following terms and definitions apply. 2.1 Total bacterial count The total number of mesophilous aerobic and facultative anaerobe colonies growing in the samples collected in air of public places after being cultivated in nutrient agar medium at 35~37℃ for 48h. 2.2 Total fungi count The number of colonies generated by the samples collected in air of public places after being cultivated in Sabouraud's agar medium at 28℃ for 5d. 2.3 β-hemolytic streptococcus The typical colony generated on blood agar plate by the sample collected in air of public places after being cultivated at 35~37℃ for 24~48h. 2.4 Legionella pneumophila The typical colony growing on GVPC agar plate through cultivation, growing on BCYE agar plate while not growing on the BCYE-CYS agar plate, which is further verified through bio-chemical experiment and serological experiment. 2.5 Impacting method
