Test guidelines on environmental safety assessment for chemical pesticides - Part 17: Trichogramma acute toxicity test 1 Scope This part of GB/T 31270 specifies the basic requirements for materials, conditions, operation, quality control, data processing, test reports, etc. for acute toxicity test of chemical pesticides on trichogramma. This part is applicable to trichogramma acute toxicity test for the registration of chemical pesticides. It may be used as reference for other types of pesticides. This part is not applicable to volatile and insoluble chemical pesticides. 2 Terms and definitions For the purposes of this document, the following terms and definitions apply. 2.1 median lethal rate application quantity of the test substance, at which 50% of the test organisms die in the acute toxicity test, expressed by LR50 Note: the unit is mg a.i./ cm2 , i.e., the amount of active ingredient of the test substance attached per unit area. 2.2 safety factor ratio of the median lethal rate LR50 of trichogramma to the recommended field applied concentration of the test pesticide, which may be expressed by Equation (1): Safety factor= (1) 2.3 test substance substance to be tested 2.4 chemical pesticide pesticide made of chemical substances through artificial synthesis, some of which are synthetic bionic pesticides made through imitation, structural transformation and innovation by taking the active substances in natural products as the matrix Synonym: synthetic organic pesticide. [Definition 2.3.1, NY/T 1667.1-2008] 2.5 technical material final product consisting of the active ingredients and impurities obtained during the manufacturing process, which shall not contain visible foreign substances or any additives, and if necessary, a small amount of stabilizer may be added [Definition 2.5.1, NY/T 1667.2-2008] 2.6 formulation product product stable in use and made of technical material (or technical concentrate) of pesticides and auxiliaries [Definition 2.4, NY/T 1667.2-2008] 2.7 active ingredient; a.i. biologically active ingredient (with a specific chemical structure) in pesticide products [Definition 3.1, NY/T 1667.2-2008] 3 Test overview Prepare the test substance into a series of diluents of different concentrations by a solvent such as acetone, quantitatively add it into a finger-shaped tube to prepare a pesticide film tube, put the test trichogramma into it to crawl for 1h and then transfer the trichogramma into a finger-shaped tube without pesticide; check the survival of trichogramma 24h later. Obtain the LR50 value and 95% confidence limit of trichogramma against the pesticide. 4 Test methods 4.1 Materials and conditions 4.1.1 Test organism Select one of the followings for test: trichogramma dendrolimi, trichogramma ostriniae, trichogramma japonicum, trichogramma evanescens, tri-chogramma confusum or Trichogramma closterae, etc. 4.1.2 Test host organism Antheraea pernyi egg or corcyra cephalonica eggs. 4.1.3 Test substance Pesticide formulation product, technical material or pure product is adopted. Dissolve the formulation with distilled water while dissolve the technical material or pure pesticide with and a solvent such as acetone. 4.1.4 Main apparatus and instruments Main apparatus and instruments are as follows: ——finger-shaped tube; ——artificial climate box; ——pipette; ——volumetric flask, etc. 4.1.5 Test conditions The test shall be performed in a dark place at a temperature of 25±2℃ and a relative humidity of 70%~80%. 4.2 Test operation 4.2.1 Pre-culture of test trichogramma Culture the host eggs in a dark place at a temperature of 25±2℃ and a relative humidity of 50%~80 % to produce trichogramma for acute toxicity test. The test trichogramma shall be derived from parasitic eggs of the same batch at the same time. Generally, most trichogrammas emerge about 24h later, thereof, those emerge within 48h are adopted for test. 4.2.2 Pre-test Arrange 4~5 concentration groups at a large dose difference according to the conditions of the formal test, and work out the maximum dose at which no trichogramma is killed and the minimum dose at which all trichogrammas are killed, so as to determine the dose range used for formal test. 4.2.3 Formal test In formal test, arrange at least five gradient concentrations (with geometric difference controlled within 2.2 times) according to the pre-test result, and set a blank control group; a solvent control group shall also be set where a solvent is used for dissolution. Arrange 3 replicates (each consisting of 100±10 trichogrammas) for the control group and each treatment group. Add a certain amount of test liquid into a finger-shaped tube, fully roll it in the finger-shaped tube until it is dried to make a film tube, put the test trichogrammas into the film tube for 1h, and then transfer them into a finger-shaped tube without pesticide, feed them with 10% honey water; then seal the tube mouth. The number of trichogrammas in the control group is the same as that in the treatment group, and both groups are tested simultaneously. Check the survival of trichogrammas in the finger-shaped tube without pesticide 24h after transferring.
